京海黄鸡促性腺激素释放激素受体基因密码子特性分析

摘  要:
为了了解鸡促性腺激素释放激素受体(gonadotropin releasing hormone receptor,GnRHR)基因的密码子特性,以便于为GnRHR基因的表达选择合适的外源表达系统,本研究使用在线工具CUSP、CHIPS及CodonW软件对鸡GnRHR基因的密码子特性进行分析,并与鸡、模式动物基因组和其他动物GnRHR基因密码子特性进行比较。结果显示,京海黄鸡GnRHR基因的ENc值较小,具有较强的密码子偏好性,且偏好以G/C结尾的密码子,GnRHR基因CDS序列中,GC含量大于AT含量;CodonW软件计算结果表明,京海黄鸡GnRHR基因密码子中,密码子GCC、ATC、CTC、CTG、CGC、CGG、AGC、TCC和GTG(RSCU>2)偏好性较强,且均以G/C结尾。对31条鸡基因分析表明鸡偏好密码子中有12种G/C结尾的密码子。与大肠杆菌、酵母菌和小鼠基因组密码子使用频率比较显示,京海黄鸡GnRHR基因密码子偏好性与3种生物相差很大,不适合进行外源表达。与其他动物GnRHR基因密码子偏好性比较显示,禽类GnRHR基因偏好含有或以G/C结尾的密码子,而其他物种的GnRHR基因无强烈的偏好性。聚类分析结果表明,亲缘关系近的物种之间倾向于拥有相同的密码子偏好性。

摘  要:
In order to understand the codon characteristics of GnRHRgene and select the appropriate exogenous expression system,online tools CUSP,CHIPS and CodonW software were used to analyze the codon characteristics of GnRHRgene.GnRHRgene was compared to other genes of chicken,model animal genomes and GnRHR gene of other species.The results showed that ENc value of GnRHRgene in Jinghai Yellow chicken was small,which indicated strong codon bias.GnRHRgene was bias toward the synonymous codons with G and C at the third codon position.GC content was greater than AT content in CDS region of GnRHRgene.Analysis by CodonW software showed that in codons of GnRHRgene in Jinghai Yellow chicken,GCC,ATC,CTC,CTG,CGC,CGG,AGC,TCC and GTG were of strong bias and end with G/C.There were 12 bias codons with G/C at the third codon position in 31 genes of chicken.Codon bias comparison to other species showed that difference between GnRHRgene and the three species were very large,which indicated that GnRHRgene was not suitable for exogenous expression.GnRHRgene of poultry bias toward the synonymous codons contained G/C or end with G/C.GnRHRgene of our species had no strong codon bias.Result of clustering based on RSCU value showed that species with close genetic relationship tend to have similar codon bias.
