Enpp2基因的分子特征及其在雌性牦牛不同繁殖阶段生殖器官中的表达

摘  要:
为了探讨胞外焦磷酸酶/磷酸酯酶2(Enpp2)基因在母牦牛不同繁殖阶段生殖器官中的表达与定位,对Enpp2基因进行克隆,并利用实时荧光定量PCR(RT-qPCR)、蛋白质免疫印迹(Western blot)和免疫组织化学法检测ENPP2在母牦牛生殖器官中的表达与定位。结果显示,Enpp2基因ORF全长2 669 bp,共编码875个氨基酸,表现为高度保守且种属间存在差异性;ENPP2蛋白是一种不稳定的可溶性蛋白,有信号肽。ENPP2在雌性牦牛生殖器官均有表达,其中在卵巢上,妊娠期的相对表达量显著高于卵泡期和黄体期(P<0.05);在输卵管上,黄体期最高,妊娠期最低(P<0.05);在子宫上,妊娠期显著低于卵泡期和黄体期(P<0.05)。免疫组化结果显示,ENPP2主要定位在黄体细胞、卵泡膜和卵泡颗粒层、输卵管黏膜上皮以及子宫内膜和子宫腺上。研究结果可为探究牦牛生殖生理提供理论基础。

摘  要:
This study was designed to investigate the expression and localization of ENPP2 gene in reproductive organs of female yaks during different reproductive stages using real-time PCR(RT-qPCR),Western blotting(WB) and immunohistochemistry techniques.Enpp2 gene was successfully cloned(GenBank accession number: MW030285).The results showed an ENPP2 gene ORF of 2 669 bp in length encoding a total of 875 amino acids.The ENPP2 protein was found to be highly conserved, varied among species; unstable, and had a signal peptide.The relative expression of ENPP2 mRNA during pregnancy was significantly higher than that during follicular and luteal stages of the ovaries(P<0.05),also, its relative expression was highest in the luteal phase and lowest during pregnancy in the oviducts.In the uterus, the relative expression of ENPP2 during pregnancy was significantly lower than that in the other two stages.ENPP2 was mainly localized in the luteal cells, theca and follicular granular layer of the ovaries, the mucosal epithelium of the oviducts, the endometrium, and uterine glands of the uterus.The results of this study provided a theoretical basis for exploring yak reproductive physiology.
