姬松茸多糖对缺氧-缺糖损伤的神经细胞线粒体膜电位和凋亡的影响

摘  要:
为研究姬松茸多糖对受损神经细胞线粒体膜电位和凋亡的影响,以大鼠海马神经细胞缺氧-缺糖再给氧为模型,以水提醇沉法自制姬松茸多糖浸膏处理细胞,应用四氮唑盐比色(MTT)法检测细胞线粒体活性,采用流式细胞仪检测凋亡细胞百分率和线粒体膜电位(MMP)的变化,并测定细胞乳酸脱氢酶(LDH)释放率。结果显示,与对照组相比,缺氧-缺糖5h后给氧,神经细胞凋亡率和LDH释放率极显著增加,并随给氧时间延长而增高,线粒体活性和MMP则极显著降低,并随给氧时间的延长而进一步下降;姬松茸多糖能明显降低细胞凋亡率和LDH释放率,提高线粒体活性和MMP,与缺氧-缺糖再给氧组相比均有显著差异。表明,姬松茸多糖可抑制缺氧-缺糖再给氧损伤所致的MMP降低,抑制细胞凋亡的发生,而对海马神经细胞发挥保护作用。

摘  要:
To study the effect of Agaricusblazeipolysaccharide on apoptosis and mitochondrial membrane potential of rat sea-horse neurocytes damaged by oxygen-glucose deprivation with the self-made Agaricus blazei polysaccharide extract by water extraction and alcohol precipitation method.The activity of mitochondria was determined by MTT test.The apoptosis percentage and mitochondrial membrane potential(MMP)changes were determined by flow cytometry.In addition,the releasing of LDH was also determined.The results showed that compared to the control group,the apoptosis percentage and release amount of LDH significantly increased when reoxygenation occured at 5hours after oxygen-glucose deprivation and increased with the prolonged reoxygention period.Activity of mitochondria and MMP significantly decreased with the prolonged reoxygenation period.The Agaricus blazei polysaccharide played effective role in lowering apoptosis percentage and release amount of LDH and raising the activity of mitochondria and MMP.It demonstrated that Agaricus blazei polysaccharide could inhibit the descent of MMP by reoxygenation after oxygen-glucose deprivation and apoptosis,and play an important role in protecting sea-horse neurocytes.
