氯霉素残留检测竞争ELISA试剂盒的研制及其性能测定

摘  要:
应用抗氯霉素单克隆抗体(CAP mAb)杂交瘤细胞株建立竞争ELISA(ciELISA)分析方法,研制出CAP残留快速检测试剂盒(CAP-Kit),并对其性能进行了测定。结果表明,CAP-Kit标准曲线呈典型的S型,符合4参数logit曲线拟合,相关系数R2=0.9955,检测范围为0.1~128.0μg.L-1,半数抑制浓度(IC50)为2.4μg.L-1,灵敏度为0.053μg.L-1,检测限为0.1μg.L-1;奶样、肉样的平均添加回收率为88.5%、82.7%,平均批内和批间变异系数均<15%;CAP-Kit与氯霉素琥珀酸钠的交叉反应(CR%)为150%,与其他酰胺醇类和抗菌药物无CR;基质对CAP-Kit的检测结果影响不大;CAP-Kit在4℃可保存6个月以上。

摘  要:
A ciELISA kit for detection of Chloramphenicol(CAPKit)was developed with generating hybridoma lines that secrete CAP monoclonal antibodies(CAP mAb) and the method of ciELISA,its traits were identified.The calibration curve of CAP-Kit was typical sigmoid curve fitted to the four parameter logistic equation with the linear detection of 0.1 to 128.0 μg·L~(-1)(R~2=0.9955),the sensitivity of 0.053 μg·L~(-1),the IC_(50) of 2.4 μg·L~(-1) and the detection limit of 1.0 μg·L~(-1).The recoveries of CAP spiked in milk were 88.5% and in pork were 82.7%.The precision and accuracy of the assay as determined by inter-assay and intra-assay coefficient variation were below 15%.The CAP-Kit had 150% cross-reactivity towards CAP succinate sodium and little or no cross-reactivity towards other amphenicols.The dilution solution of CAP had no effects on results of CAP-Kit.The validity of CAP-Kit in 4℃ was above six months.
