二倍体苜蓿扦插扩繁、移栽及自交结实率分析

摘  要:
在前期研究基础上,以四倍体组培苗为对照,利用组培扦插的方式扩繁苜蓿单倍体植株,筛选适合单倍体炼苗移栽的营养土,并对移栽成活的单倍体植株进行自交授粉,观测其生长和结荚情况。结果显示,新疆大叶苜蓿二倍体植株组培扦插适宜的生根培养基为1/2 MS+0.1mg/L NAA+2%蔗糖+0.7%琼脂,生根率可达83%。进行炼苗移栽时,采用无菌营养土可使二倍体成活率达41.7%。苜蓿二倍体植株自交结实率和种子活力指数均显著高于四倍体。

摘  要:
In order to rapidly propagate haploid plants and solve the problem that it is difficult for alfalfa haploid to survive after transplanting. On the basis of previous research, the tetraploid tissue culture seedlings of alfalfa were used as the control, the haploid were propagated by tissue culture cutting, the nutrient soil suitable for haploid hardening-seedling transplanting was screened, and the transplanted haploid plants were self-pollinated to observe their growth and pod fermentation. The results showed that the suitable rooting medium for diploid cuttings of Medicago sativa L. cv. Xinjiang Daye was 1/2 MS+0.1 mg/L NAA+2% sucrose+0.7% agar, and the rooting rate was 83%. When the seedlings were transplanted, the survival rate of diploid plants could reach 41.7% by using aseptic nutrient soil. The inbred pod setting percentage and seed vigor index of alfalfa diploid plants was significantly higher than that of tetraploid plants.
