基于Illumina高通量测序的马来熊(Helarctos malayanus)粪便菌群多样性初探

摘  要:
应用新一代高通量测序技术Illumina Miseq对上海动物园3只马来熊的12个粪便样本中的菌群结构及其多样性进行检测和分析。样中共检测到884 522条序列,共检测到15个菌门,主要有7个优势菌门,按照多样性高低依次是:厚壁菌门(Firmicutes)(54. 70%)、变形菌门(Proteobacteria)(41. 90%)、梭杆菌门(Fusobacteria)(1. 80%)、蓝藻菌门(Cyanobacteria)(0. 70%)、放线菌门(Actinobacteria)(0. 20%)、拟杆菌门(Bacteroidetes)和未定门(TM7)。粪便样本中能够确定到菌属的序列占73. 10%,以链球菌属(Streptococcus),大肠杆菌/志贺菌属(Escherichia-Shigella)和梭状杆菌属(Clostridium)为优势菌属。3只马来熊粪便菌群多样性比较:LS02粪便菌群多样性最高,其次是LS01,和LS03。结论:马来熊粪便菌群存在个体差异,但同一个体的优势菌群组成非常相似,年龄和性别可能会对马来熊粪便菌群产生影响。

摘  要:
We collected 12 fecal samples of 3 sun bears Helarctos malayanus( LS01,LS02 and LS03) at Shanghai Zoo,and examined and analyzed the fecal macrobiotic structure and diversity using Illumina Miseq. In total,884 522 sequences were found in the fecal samples,and 15 bacteria phyla were identified. The seven predominant phyla were Firmicutes( 55%),Proteobacteria( 42%), Fusobacteria( 2%), Cyanobacteria( 0. 7%), Actinobacteria( 0. 2%), Bacteroidetes and TM7. Of these sequences,73% were identified to genus,and Streptococcus,Escherichia/Shigella and Clostridium were predominant. Sun bear number LS02 showed the highest fecal bacterial diversity,followed by LS01 and LS03 in descending order. There were individual differences among fecal microbe composition,but the predominant bacteria classification was similar.Sex and age might impact the fecal microbe composition of H. malayanus.
