家蚕胚胎期单卵基因组DNA的快速抽提

摘  要:
本文对用磁珠法抽提家蚕单粒蚕卵基因组DNA进行了探讨。结果表明从催青24h后到收蚁前一天的单卵中所提取的DNA断裂少、纯度高,能够满足特异性引物或随机引物进行PCR扩增的需要。每粒卵可获得100ng以上的DNA,能够进行多次重复检测,完全能够满足蚕种鉴定及纯度检测的需要。

摘  要:
This article is about the method of magextracting genomic DNA from single silkworm egg. We could extract genomic DNA from a single silkworm egg in the period from 24h to the day before hatching during egg incubation. The extracted genomic DNA has less breakage with high purity, and meet the requirements for PCR amplification. From every extracted silkworm egg we could get more than 100 ng genomic DNA that is enough for identification of silkworm breed for the evaluation of its purity.
