双抗可促进子宫颈细胞天然防御反应——DEFA1和DEFB

摘  要:
为了研究抗生素对子宫颈细胞天然防御(防御素)的影响,试验使用1%青霉素和1%链霉素处理Hela细胞,检测DEFA1和DEFB1的表达;使用不同浓度的双抗处理Hela细胞,检测DEFA1和DEFB1的表达;使用无双抗培养液培养Hela细胞,在不同时间检测DEFA1和DEFB1的表达。结果表明:双抗能促进Hela细胞防御素的表达,随着双抗浓度从1%增加到5%,DEFA1和DEFB1表达量显著升高(P <0. 05)。在抗生素刺激消失的72 h内,mRNA水平、DEFA1和DEFB1表达量显著下降(P <0. 05),蛋白水平DEFA1表达量不变(P>0. 05)。说明青霉素和链霉素这两种抗生素能促进Hela细胞产生防御素DEFB1;抗生素除了有直接杀菌作用外,还通过刺激子宫颈细胞产生防御素间接杀菌。

摘  要:
Cervical epithelial cells( Hela cells) were treated with penicillin and streptomycin to evaluate the effect of antibiotics on production of two natural defenses in cervical epithelial cells. Method: Hela cells were treated with 1%penicillin and 1% streptomycin to detect the expression of DEFA1 and DEFB1. Hela cells were treated with different concentrations of penicillin-streptomycin mixture to detect the expression of DEFA1 and DEFB1. Hela cells were cultured without Penicillin-Streptomycin for different time. Results: two antibiotics can promote the expression of defensins in Hela cells. The expression of DEFA1 and DEFB1 increased significantly with the increase of the concentration of two antibiotics from 1% to 5%( P < 0. 05). Within 72 hours after antibiotic stimulation disappeared,at the level of mRNA,the expression of DEFA1 and DEFB1 decreased( P < 0. 05),and at the level of protein,the expression of DEFA1 have not changed( P > 0. 05). Conclusion: Both penicillin and streptomycin can promote the production DEFB1 in the cervical epithelial cells.
