黄麻链霉菌AUH-1抗菌活性物质发酵优化研究

摘  要:
黄麻链霉菌AUH-1具有广谱拮抗作用,能产生对水稻纹枯病菌、西瓜枯萎病菌等多种植物病原真菌具有强烈抑制作用的活性物质。采用响应面法对黄麻链霉菌AUH-1培养基成分中的碳源、氮源进行优化,以期提高黄麻链霉菌AUH-1对水稻纹枯病菌的抑制效果。利用不同碳氮源进行单因素实验,根据单因素实验结果确定碳氮源水平,应用Design-Expert 8.05b软件中Box-Benhnken设计响应面试验,得到黄麻链霉菌AUH-1发酵培养基的最优配比组合浓度。结果表明,优化后培养基的成分浓度为蔗糖3.1%、玉米淀粉0.25%、玉米浆4.5%、硫酸铵0.375%,在此浓度配比下黄麻链霉菌AUH-1对水稻纹枯病菌的抑制效果最佳,模型预测值为70.504 1%,实际优化值达到69.31%,相比基础培养基提高了5.78%。为进一步研制水稻纹枯病生防制剂和杀菌剂奠定基础。

摘  要:
Streptomyces corchorusii AUH-1 had broad-spectrum in suppressing the growth of various fungal plant pathogens, such as Rhizoctonia solani and Fusarium oxysporum f.sp.niveum. In order to improve the inhibitory effect of S. corchorusii AUH-1 on R.solani, the response surface methodology was used to optimize the carbon source and nitrogen source in the culture medium in this paper. A single factor experiment was designed and tested with different concentration of carbon source and nitrogen source. According to the test results, the carbon source and nitrogen source were selected as variables. Design-Expert 8.05 b Software was used in the design of the test group of Box-Benhnken, the data were analyzed by using Design-Expert 8.05 b software, and the optimal combination concentration of carbon source and nitrogen source was obtained. The results showed that the optimal component level of each factor was 3.1% sucrose, 0.25% corn starch, 4.5% corn steep liquor and 0.375%ammonia sulfate, and the inhibitory effect of S. corchorusii AUH-1 on R.solani was the best under this concentration. The model prediction value was 70.5041% and the actual optimized value was 69.31%, which was5.78% higher than that of the basic medium. This study laid a foundation for the further development of rice sheath blight bio-control agents and fungicides.
