摘 要:
研究旨在检测低氧诱导因子2α基因(Hypoxia inducible factor-2α,HIF-2α)和血管内皮生长因子基因(Vascular endothelial growth factor,VEGF)在14胚龄藏鸡不同组织中的差异表达情况及两基因在不同组织中表达的相关性。低氧条件下孵化藏鸡种蛋至14胚龄时,取鸡胚肝脏、胸肌、腿肌、卵黄膜、尿囊膜、下丘脑6个组织;使用试剂盒从上述组织中提取总RNA,反转录后对各组织中HIF-2α基因、VEGF基因及内标β-actin基因进行实时荧光定量PCR检测;利用统计软件分析HIF-2α和VEGF基因在不同组织中的表达是否有差异及两基因相对表达量之间是否具有相关性。结果显示,HIF-2α基因在尿囊膜中表达最高,肝脏次之,其余四个组织中HIF-2α基因表达量较低且彼此间无显著差异。肝脏中出现VEGF基因特异性高表达,其余组织中VEGF基因表达量由高到低依次为腿肌、胸肌、尿囊膜、卵黄膜、下丘脑。尿囊膜组织中HIF-2α基因与VEGF基因表达量呈负相关,其他组织中两基因表达均不相关。研究发现尿囊膜和肝脏中HIF-2α基因高表达以及VEGF基因在肝脏中的高表达可能是藏鸡早期发育过程中应答低氧环境的重要机制之一。HIF-2α基因与VEGF基因在尿囊膜组织中表达呈负相关是藏鸡适应低氧环境的表现。

摘 要:
The study was conducted to detect the relative expressions of hypoxia inducible factor-2 alpha(HIF-2α)gene and vascular endothelial growth factor(VEGF)gene in different tissues of Tibetan chicken embryos at embryonic age of 14 days and whether they had correlation in different tissues. Tibetan chicken breeding eggs were hatched under the hypoxia circumstance for 14 days,then took the tissues of liver,pectorales,gastrocnemius,vitelline membrane,chorioallantoic membrane and hypothalamus from the embryo. The total RNA of above tissues was extracted by kit.The real time PCR technique was used to test the expressions of HIF-2α gene,VEGF gene and β-actin gene(internal standard)in above tissues after reverse transcription. The differential expression of HIF-2α gene and VEGF gene and their correlation in different tissues was analyzed by statistical software. As a result,HIF-2α gene had the highest expression in chorioallantoic membrane tissue and had the second highest expression in liver tissue,the expression of HIF-2α gene was lower in other four tissues and there were no significant differences between each other. The highest expression of VEGF gene was observed in liver tissue,the sequence of VEGF gene expression from high to low in other tissues was gastrocnemius,pectorales,chorioallantoic membrane,vitelline membrane,hypothalamus. There was a negative correlation between HIF-2α gene and VEGF gene in chorioallantoic membrane tissues,and there was no correlation in other tissues. In this study,we indicated that the high expression of HIF-2α gene in chorioallantoic membrane,liver and the high expression of VEGF gene in liver may be one of the important way for Tibetan chicken to reply the hypoxia environment during early development. The negative correlation between HIF-2α gene and VEGF gene in chorioallantoic membrane tissues was the representation for Tibetan chicken to adapt the hypoxia environment.
