紫云英根瘤菌的质粒特征及内源抗药性

摘  要:
对22个野生型紫云英根瘤菌菌株的质粒和抗性基因的研究结果表明,不同紫云英根瘤菌菌株都含有1-4个数量不等的质粒,其分子量变幅在18.5-300Kb之间。大部分野生型分离菌具有不同程度的抗庆大霉素,卡那霉素,四环素和链霉素的能力;其中抗庆大素和抗卡那霉素的能力超出一般快生型根瘤菌的许多倍。对菌株HR104的热处理消除试验结果表明,抗庆大霉素的因子与其中的一个巨型质粒(p104c)相关。用消除质粒p104c的突变株回接寄主植物仍能够形成有效根瘤,说明该质粒的存在与共生固氮过程无关。

摘  要:
Plasmids and endo-antibiotic resistance were investigated among 22 wildstrains of R. astragalii. The investigated strains contain variably 1-4 plasmids of 18. 5 to300Kb. Most strains are resistant to gentamycin (Gm, 20-100μg/ml), kanamycin (km,50-200μg/ml),tetracycline(Tc, 5-30μg/ml)and streptomycin(Sm,5-100μ/ml)£(r)TheGm and Km resistances are many times higher than those of other fast-growing Rhizobiumspecies. By heat curing treatment of strain HR104, 8 Gm sensitive mutants were selected,among which one mutant lost its largest plasmid p104c, two mutants shweod deletion ofp104c. plant inoculation experiment showed that mutants with loss or deletion of p104ccould induce effective nodules on Astragalus sinica, indicating that the plasmid p104c isnot responsible for the symbiotic process.
