云南昭通苹果产区苹果花叶病毒的鉴定

摘  要:
研究通过电镜、免疫捕获RT-PCR的方法,对云南昭通表现花叶的苹果病样进行鉴定,电镜观察到直径为26~35 nm,无包膜二十面体的准等轴对称病毒颗粒,与苹果花叶病毒(Apple mosaic virus,ApMV)相似。根据苹果花叶病毒的外壳蛋白基因序列设计、合成引物,提取病毒病样品总RNA,进行扩增、克隆和测序。序列分析表明侵染云南昭通的苹果花叶病病原为ApMV。把本试验鉴定的苹果花叶病毒与已报道的31个苹果花叶病毒分离物进行序列比对和分析。所有的ApMV分离物可以划为3个亚组。其中,亚组Ⅰ、亚组Ⅱ中均有侵染苹果的ApMV分离物。亚组Ⅰ寄主分化较多,亚组Ⅱ多为苹果分离物,而梨分离物单独聚在亚组III。本研究的云南昭通ApMV分离物(ZT)与昆明斗南月季分离物(Kmi2),美国分离物(NCGR9026)、捷克共和国分离物4个分离物(Cz1,Cz2,Cz3,Cz5)、德国分离物(G1)聚为寄主分化较多的亚组Ⅰ,与昆明斗南月季分离物Kmi2的核苷酸序列同源性最高,相似性达到99.8%。本研究为研究云南ApMV分子变异、与寄主的互作和指导防控提供依据。

摘  要:
In this study,the disease samples showed mosaic symptom were collected in Zhaotong,and detected by electron microscope.The particles with 26-35nm diameters were observed under electron microscope,which similar to the characters of Apple mosaic virus(ApMV).The total RNA was extracted from infected apple leaves and the specific primers were designed for RT-PCR.Immunocapture RT-PCR was conducted and PCR products were cloned and sequenced.The sample showing mosaic symptom was identified an isolates of ApMV.The sequence of ApMV Zhaotong isolate(ZT) was compared with 31 isolates ApMV.The results showed that all the isolates were grouped into three clusters.The isolates infecting apple were grouped into cluster Ⅰand cluster Ⅱ.The isolates of cluster Ⅰ come from different hosts.However,most isolates of cluster Ⅱ come from apple.And all the isolates of cluster III come from pear.ApMV Zhaotong isolate(ZT),Kunming Dounan isolate(Kmi2),the United States isolate(NCGR9026),four isolates from Czech Republic(Cz1,Cz2,Cz4,Cz5) and Germany isolate(G1) were grouped into clusterⅠ,which showed varied hosts.The nucleotide sequence of CP gene of ApMV Zhaotong isolate(ZT) was the nearest to Kmi2 isolate from Kunming Dounan,which showed 99.8% nucleotide similarities.The studies provide the basis that study molecular variation in Yunnan,host interactions and resistance and control to apple mosaic diseases.
