高温胁迫对荒漠植物花花柴大分子结构的影响

摘  要:
高温胁迫会损伤植物细胞,严重时使核酸及蛋白质发生降解。试验采用45℃高温处理花花柴植株0 h、2 h、4 h、8 h、12h,分别提取不同处理时间叶片DNA、RNA及蛋白质进行电泳,分析其降解程度;同时利用彗星电泳分析不同处理时间细胞的损伤程度。结果表明,花花柴叶器官经45℃处理12 h时DNA明显降解,处理8 h后RNA开始降解且蛋白质降解明显。通过彗星电泳检测发现处理8 h后花花柴叶片细胞变形、明显受损。表明花花柴耐受45℃高温的临界时间为8 h,其具有极强的耐高温性,可作为研究植物耐高温的重要材料和高温荒漠区生态恢复及重建的重要种质资源。同时,通过电泳检测DNA、RNA及蛋白质的降解程度,为评估细胞损伤程度提供了一个简单而有效的评价方法。

摘  要:
The cell, nucleic acids and proteins of plant are destroyed under high temperature stress. In order to explore the integrity of DNA, RNA, protein, and cell of Karelinia caspia under high temperature condition, the DNA, RNA and protein were extracted and analysed by gel electrophoresis at 0, 2, 4, 8, 12 h under 45 ℃ treatment, respectively. Meanwhile, the cellular integrity was detected using comet assay. Result showed that the DNA structure was destroyed at 12 h and the RNA was damaged at 8 h, and the protein was obviously degraded at 8 h under 45 ℃. The cell was severely damaged and deformed at 12 h under 45 ℃ by comet assay. It showed that the critical time was 8 h for Karelinia caspia under 45 ℃. The Karelinia caspia has very strong resistance to high temperature stress and self-adjustment adaptability. It could be used as an important plant resources to research characteristic of resistance to adversity, and as plant resources used ecological restoration and reconstruction in desert area. At the same time, it provides a simple and effective method to evaluate the degree of cell damage.
