薤白EPSPs基因在不同组织表达的半定量分析

摘  要:
5-烯醇式丙酮酸-3-磷酸合成酶(EPSPs)是植物芳香族氨基酸合成途径的重要酶,是优良除草剂草甘膦作用的靶酶.根据已克隆的抗草甘膦植物薤白EPSPs基因cDNA序列的3′端非保守区域设计引物,采用RT-PCR半定量技术,研究了该基因在薤白不同组织中的表达情况.利用18S rRNA为表达的内参基因,建立一个针对EPSPs特异、稳定的RT-PCR半定量检测体系.对EPSPs基因表达的检测表明,薤白幼叶中基因表达水平最高,总表达水平高低依次为:幼叶,根,茎,壮叶.相对18S rRNA表达量为:幼叶0.631,根0.246,茎0.218,壮叶0.120.

摘  要:
5-enolpyruvlshimimate-3-phosphate synthase (EPSPs) is a key enzyme catalyzing the synthesize of aromatic amino acids (shikimate pathway) in plant.It is the target enzyme of a wide used herbicide glyphosate.A stable and specific RT-PCR semi-quantitative system for EPSPs expression detection is settled with a pair of specific primers designed according to the 3′ non conservation region of the cloned EPSPs gene cDNA of Allium macrostemon Bunge,a glyphosate tolerance weed.The 18S rRNA is used as internal control for the expression test.The quantitative result shows that the EPSPs expression in Allium macrostemon Bunge is at the highest level in young leaves and less in old leaves.The expression model is in a sequence of young leaves,root,stem,old leaves from higher to lower. The relative expression level of EPSPs to 18S rRNA is 0.631,0.246,0.218 and 0.120 respectively.
