基因工程菌生产耐高温α-淀粉酶发酵条件的优化

摘  要:
利用重组毕赤酵母基因工程菌诱导表达可产生耐高温α-淀粉酶,采用摇瓶发酵,从诱导时间、甲醇添加量、pH值、接种量4个因素考察了发酵条件对耐高温α-淀粉酶酶活的影响,结合四元线性回归正交设计研究方法构建出甲醇诱导重组毕赤酵母生产耐高温α-淀粉酶的回归模型;优化最佳工艺参数为:诱导时间96h、甲醇添加量0.5%、接种量150mL、摇床转速200rpm;最优工艺条件下,耐高温α-淀粉酶酶活为217.2U/mL。

摘  要:
The recombinant Pichia pastoris genetic engineering bacteria is used to produce thermostable α-amylase.By the shaking flask,the influences of inducing time,methanol,pH and immunization rate on the expression of thermostable α-amylase which is measure from the active of thermostable α-amylase were investigated.Four-linear regression with orthogonal design methods construct recombinant Pichia pastoris methanol induction of α-amylase production of thermostable regression model.The optimization of fermentation condition is that inducing 96h,adding 0.5% methanol,inoculation 150mL,shaking revolution 200rpm.Under this optimum conditions,the thermostable α-amylase activity was 217.2U/mL.
