羊骨蛋白酶解物免疫活性及酶解条件的研究

摘  要：
【目的】利用羊骨中胶原蛋白以获得免疫调节活性肽。【方法】利用木瓜蛋白酶对羊骨蛋白进行酶解,采用四元二次通用旋转设计,考察酶用量(E/S)、水解温度(Temp)、水解时间(t)和底物浓度(S)对免疫活性的影响,并选出最佳的酶解条件。同时采用Folin-酚法测定了酶解产物中可溶性肽的含量,采用SAS软件分析可溶性肽含量与免疫活性之间的相关关系。经Design expert7.1.2软件优选最佳酶解条件。【结果】得出木瓜蛋白酶的最佳水解条件为:酶用量(E/S)1576U·g-1,水解温度为(Temp)64.05℃,水解时间(t)7.22h,底物浓度为(S)0.271kg·L-1。可溶性肽含量与免疫活性之间的相关系数r=0.045

摘  要：
【Objective】 Utilizing livestock processing waste sheep bone to get immunocompetence peptide.【Method】Enzymatic Hydrolysis conditions were optimized using a four elements twice general rotation design method. The temperature (Temp), time (t), enzyme/substrate (E/S) ratio and substrate concentration (S) on the amount of TCA soluble peptides were considered. The content of soluble peptide in the acquisition was measured by Folin-hydrozybebzebe method, and the relation of soluble peptide content with immunocompetence was analyzed by SAS software. The best enzymatic hydrolysis condition was gotten from Design Expert 7.1.2 software. 【Result】The condition of enzyme/substrate level of 1 576 U·g-1, temperature of 64.05℃, substrate concentration of 0.271 kg·L-1 and a hydrolysis time of 7.22 hours is the optimum condition to obtain a higher immunocompetence acquisition. The correlation coefficient between TCA soluble peptides and the immuneocompetence is 0.045,that is less than r0.05=0.355, showing that they had nosignificant correlation.【Conclusion】 The result showed that the protein is mainly composed of peptides, but it’s content do not play a important role on the immunocompeptence.
