河西走廊14种锦鸡儿遗传多样性SSR分析

摘  要:
选用9对细胞核SSR(nSSR)和10对叶绿体SSR(cpSSR)标记,对来源于甘肃省民勤沙生植物园的14种锦鸡儿(Caragana Fabr.)进行遗传多样性分析。9对nSSR引物共检测到109个多态位点,引物的多态性信息量(PIC)变动在0.320~0.921之间,平均0.738;10对cpSSR引物共检测到92个多态性位点,PIC变动在0.512~0.865之间,平均0.671;14种锦鸡儿种间遗传相似系数在0.60~0.92之间,平均0.76;聚类分析将14种锦鸡儿划分为5大类群6个亚类,nSSR和cpSSR所分析的种间遗传关系与传统的分类结果基本一致;分子标记结果表明,锦鸡儿属植物具有丰富的遗传多样性,为锦鸡儿属植物种质资源及育种利用提供理论依据。

摘  要:
The genetic diversity and relationship were investigated using 9 pairs of nSSR(nuclear simple sequence repeat) and 10 pairs of cpSSR(chloroplast simple sequence repeat) markers among 14 Species of Caragana Fabr.collected from Minqin Desert Botanical Garden of Gansu Province.Total 109 polymorphic loci were detected by 9 pairs of nSSR primers and the value of allelic PIC(polymorphism information content) ranged from 0.320 to 0.920,averaged 0.738 per primer.Total 92 polymorphic loci were detected by 10 pairs of cpSSR primers and the value of allelic PIC ranged from 0.512 to 0.865,averaged 0.671 per primer.The value of genetic similarity(GS) indexes among those 14 species varied from 0.60 to 0.92 and averaged 0.76.Cluster analysis classified those 14 Species into 5 groups and 6 sub-groups.The interspecific genetic relationship analyzed by nSSR and cpSSR was almost consistent with the result by traditional classification method.The results of molecular markers assay indicated that the Caragana Fabr.had a high level of genetic diversity which could be theoretical basis for the germplasm utilization and breeding of Caragana Fabr.
