氯霉素胶体金快速检测试剂条的研制及在水产品中的应用

摘  要:
应用胶体金免疫层析技术,研制一种水产品中氯霉素免疫胶体金快速检测试剂条。将制备的金溶液以最适蛋白标记量标记抗氯霉素单克隆抗体制成金标抗体,将羊抗鼠IgG、氯霉素-牛血清白蛋白(CAP-BSA)和金标抗体三者经过反复调试优化,以适宜质量浓度和包被量分别包被到硝酸纤维素(NC)膜和胶体金结合垫上。NC膜经37℃恒温干燥32h或25℃恒温干燥7d后,和样品垫、胶体金结合垫、吸水垫及其他辅料组装成试剂条。经实验,该试剂板对水产品中氯霉素的检测限为0.3μg/kg,假阳性率4%,假阴性率为2%,常温保质期6个月以上。

摘  要:
A colloidal gold test strip for rapid detection of chloramphenicol in aquatic products has been developed and applied.Colloidal gold particles were prepared and labeled to an anti-chloramphenicol(CAP) monoclonal(MAb) antibody.Optimum concentrations of goat anti-mouse IgG,chloramphenicol-bovine serum albumin(CAP-BSA) and the gold labeled antibody were established by repeated testing before coating a nitrocellulose(NC) membrane and a colloidal gold conjugate pad.After being dried at 37 ℃ for 32 h or 25 ℃ for 7 d,the NC membrane,together with a sample pad,a colloidal gold conjugate pad,a absorbent pad and other accessories,was used to fabricate a colloidal gold test strip.The detection limit of the test strip for chloramphenicol was 0.3 μg/kg,and false positive rate(FPR) and false negative rate(FNR) were 4% and 2% respectively.Its period of validity was more than 6 months at room temperature.
