紫草愈伤组织培养中紫草素形成的影响因素

摘  要:
采用两阶段培养法研究不同培养基、激素的种类、浓度和各种营养成分对愈伤组织生长及紫草素形成的影响。结果表明 ,改良B5培养基为适宜的营养生长培养基。细胞分裂素和生长素均为紫草素形成所必需的。 6 BA 1 0mg·L- 1,IAA 0 1mg·L- 1为较佳配比。生长培养基中CaCl2 浓度为 2 5 0mg·L- 1,MgSO4 为 5 0 0mg·L- 1时 ,愈伤组织生长和紫草素形成均较佳。紫草素形成培养基中CuSO4 为M9浓度的 3倍时 ,紫草素产量较高。紫草素形成培养基中添加VB2及更高的FeSO4 对紫草素产量的提高没有效果。研究过程中优化了紫草细胞培养形成紫草素的方法 ,为建立完整的组培生产紫草素体系提供了基础数据

摘  要:
Two stage culture method was used to study the effect of types and concentration of hormone,effect of types of culture medium,and nutrition on shikonin formation.The results show that both cytokinin and auxin are needed in shikonin formation.6 BA 1 0 mg·L -1 and IAA 0 1 mg·L -1 is suitable in shikonin formation medium.B 5 medium is a better medium for cell growth.B 5 with 250 mg·L -1 of CaCl 2,500 mg·L -1 of MgSO 4 are better for both growth and shikonin formation.M9 with three times of concentration of CuSO 4 are better choice for shikonin formation.Addition of VB 2 and more FeSO 4 has no positive effect on shikonin formation.
