松江鲈脾组织细胞的短期培养及染色体分析

摘  要:
采用组织块法对体质量为120~140 g的松江鲈Trachidermus fasciatus脾组织细胞进行短期培养并对染色体进行了分析。结果表明:在p H为7.2、温度为25℃的条件下,松江鲈脾组织细胞在含有20%南美胎牛血清(FBS)、人碱性成纤维样细胞生长因子(b FGF)、硫酸软骨素、Ⅰ型胰岛素样生长因子(IGF-Ⅰ)的培养基中,生长良好,迁出细胞有悬浮和贴壁两种形式;悬浮细胞为圆形和椭圆形,贴壁细胞呈变形细胞样和成纤维细胞样;利用悬浮细胞进行的染色体分析显示,松江鲈的染色体数2n=40,核型公式为14m+10sm+16t。本研究结果为松江鲈脾组织细胞系的构建奠定了基础,并初步建立了利用短期培养的脾组织细胞制备染色体的方法。

摘  要:
Primary culture of spleen and karyotype analysis wre performed in roughskin sculpin Trachidermus fasciatus with body weight of 120- 140 g by a tissue explanting method. The results showed that the optimal growth of spleen cells in roughskin sculpin was observed in the culture medium of DMEM / F12 supplemented with 20% FBS of basic fibroblast growth factor( b FGF),insulin- like growth factor- Ⅰ( IGF- Ⅰ) and chondroitin sulfate at 25℃ and p H 7. 2. There were two types of growing spleen cells,round and oval suspended cells and cell- like and fibroblast- like adherent cells. The analysis of suspended cells revealed that roughskin sculpin had a diploid chromosome number of 40,with karyotype formula 2n = 40,and 14 m + 10 sm + 16 t. The findings provided a useful foundation for the establishment of in vitro cell- line and the functional investigation in roughskin sculpin spleen,and the chromosome preparation method was preliminarily established using cells of the spleen in roughskin sculpin.
