苹果茎痘病毒基因变异及重组分析

摘  要:
苹果茎痘病毒(apple stem pitting virus,ASPV)是一种严重危害果树生产的潜隐性病毒,为了解ASPV的变异特点,基于已报道的ASPV外壳蛋白(coat protein,CP)基因核苷酸序列信息,结合RNA二级结构分析,对ASPV的变异进行分析。结果显示,不同分离物CP基因前半部分区域存在3个大段的空位区。中国分离物PR1在CP基因末端有12个核苷酸的序列插入,RNA二级结构预测显示,插入点附近能形成一个稳定的茎环结构,序列插入并未引起RNA二级结构的明显变化。重组分析显示,波兰分离物ST181为中国分离物WS和波兰分离物N1的重组体,系统进化分析支持该重组事件的发生。ASPV核苷酸序列变异复杂,并能够发生重组,因而有必要收集更多抗性资源,加强抗病育种。

摘  要:
Apple stem pitting virus(ASPV)is one of the most important latent viruses that infect fruit trees.In order to understand the variation character of ASPV,the variation of ASPV was analyzed in the study on the basis of the nucleotide sequences of coat protein(CP)gene of various isolates and RNA secondary structure.The results indicated that there were three large blank regions in the frontal region of CPgene for various isolates.There was a 12-nt insertion in the terminal of CPgene for Chinese isolate PR1.In addition,a stable stem-loop structure could be formed for the sequences near the place of insertion,and the RNA secondary structure did not show distinct change on account of the insertion.Recombination analysis showed that the isolate ST181was a product of recombination between isolate WS from China and isolate N1from Poland,and the recombination was also supported by phylogenetic analysis.To our knowledge, this is the first report for recombination of ASPV.The variation of ASPV is complicated and recombination of genome is also found,so more anti-ASPV resources need to be collected and the anti-ASPV breeding needs to be strengthened.
