齐墩果酸对食管癌细胞Eca-109的增殖抑制作用

摘  要：
为了研究齐墩果酸(OA)在体外对食管癌Eca-109增殖的抑制作用,用不同浓度的OA对体外培养的食管癌Eca-109细胞分别干预24、48、72h后,通过显微镜观察Eca-109细胞的形态学改变,采用MTT法观察OA对Eca-109细胞增殖的抑制作用。结果表明:(1)与阴性对照组相比,5～40μg/mL浓度的OA对Eca-109细胞增殖未表现出抑制作用;80～160μg/mL浓度的OA分别作用于Eca-109细胞24、48、72h后,OA抑制Eca-109细胞的增殖,且呈一定的剂量-时间效应关系,生长抑制率最高可达87.9%。(2)浓度和作用时间对Eca-109细胞增殖均有抑制作用,且两者存在交互作用;综合时间抑制曲线,在时间为48h,剂量为160μg/mL是达到时间-剂量最优组合,即此时达到对食管癌Eca-109细胞最大抑制作用。(3)在显微镜下观察,用药组的细胞生长明显被抑制,胞浆内颗粒增多增粗,细胞形态变化明显,部分细胞变圆悬浮于培养基中。由此可知,OA在一定浓度范围内能明显抑制Eca-109细胞的增殖。

摘  要：
To observe the inhibiting effect of oleanolic acid(OA) on esophageal cancer Eca-109 cell in vitro.Esophageal cancer Eca-109 cells were cultivated in vitro and interfered by different concentrations of oleanolic acid for 24,48 and 72 hours respectively.The inhibiting effect of OA on Eca-109 cells was detected with MTT assay and microscope.1) Eca-109 cell kept proliferating in 5-20μg/ml concentration.Compared with negative control group,40-160μg/ml concentration of OA had effect on esophageal cancer Eca-109 cell after 24,48 and 72 hours respectively,and there was dose-time effect relation between OA and esophageal cancer Eca-109 cell proliferation.In addition,the maximum growth inhibition ratio reached to 87.9%.2) OA had an inhibiting effect on the esophageal cancer Eca-109cells.At the same time an interaction effect between time and dose was observed.When the intervention time was 48 hours and the dose was 160μg/ml,the dose-time was optimal combination.3) Under microscope,it's obvious that the cell growth of pharmacy group had been inhibited.Meanwhile,the number and size of cytoplasma granulae increased and the cell morphology had clearly changed;a part of cells became round,and suspended in the culture medium.Conclusion:OA has distinct inhibitory action on esophageal cancer Eca-109 cell proliferation in a certain concentration range.
