加拿大一枝黄花无菌苗培养体系建立方法研究

摘  要:
为了避免土壤中植物根系提取物定性定量分析的复杂性和不确定性,在研究加拿大一枝黄花入侵过程的化学生态机制项目工作中,建立加拿大一枝黄花无菌苗培养体系是一个必不可少的步骤。研究通过正交试验设计原理,筛选了无菌苗灭菌条件、植物激素浓度水平变化对无菌苗生长参数的影响和温度对无菌苗生长系数变化等条件参数。试验结果表明,2.0%NaClO灭菌90 s处理对加拿大一枝黄花水培苗灭菌效果最好;基于对无菌苗生长长度指标分析可以看出,2、4-D浓度的变化对其影响最大,IAA次之;基于对无菌苗生长分蘖数这个指标分析可以看出,2、4-D浓度的变化对其影响最大,IAA次之;基于对无菌苗生长叶片数这个指标分析可以看出,IAA浓度的变化对其影响最大,NAA次之。添加6-BA 5.0mg/L、NAA 0.5 mg/L、2、4-D1.0 mg/L、IAA 0.1mg/L的培养基在36~30℃的变温条件下培养无菌苗表现茎粗壮,叶片较多,生长速率快,可为研究加拿大一枝黄花入侵过程的化学生态机制项目工作提供较好的实验材料。

摘  要:
In order to avoid complexity and uncertainty of the qualitative and quantitative analysis of plant roots extracts in soil,the establishment of Solidago canadensis sterilization culture system is an indispensable step while it researches the chemical ecology mechanism of Solidago canadensis invasion process.Through orthogonal design principle,the sterilizing conditions,relationship between changes of plant hormone concentrations level and growth parameters of seedling and temperature effect on the growth coefficient parameters of seedling were screened.The results showed that the impact of sterilized way for 90 seconds by 20%NaClO on the seedling culture of Solidago canadensis was the best.Based on the length indicator of seedling,it could be seen that 2,4 - D concentration changes had the greatest impact;Based on the tillers number indicator of seedling,it could be seen that 2,4 - D concentration changes also had the greatest impact;Based on the leavers number indicator of seedling,it could be seen that IAA concentration changes had the greatest impact;Adding 6 - BA 5.0mg / L, NAA0.5 mg/ L,2,4-D 1.0 mg/ L,IAA 0.1 mg / L into culture medium under 36℃-30℃temperature conditions,the growth performance of the sterilized seedling was sturdy stems,more leaves,faster growth rate.The seedling could be used in next experiment.
