食淀粉乳杆菌对轮状病毒受体HSC70的影响

摘  要:
旨在探究食淀粉乳杆菌(Lactobacillus amylovorus)对轮状病毒(rotavirus,RV)的细胞受体热休克同源蛋白HSC70的调节作用。以饲喂食淀粉乳杆菌的中国昆明鼠乳鼠为试验动物,通过ELISA、qPCR、冰冻切片间接免疫荧光等技术,1~11 d逐日进行空肠组织取样,检测食淀粉乳杆菌对小鼠空肠组织中HSC70调节作用的动态变化。试验分为4组:正常组、病毒组、预防组(先益生菌作用后攻RV)、治疗组(先攻RV后作用益生菌)。试验结果显示,乳鼠体质量的增长率及变化情况与RV的感染相关,并且食淀粉乳杆菌可以显著地治疗及预防RV感染(P<0.05)。对空肠组织中HSC70的检测发现,攻RV后正常组及预防组的热休克同源蛋白HSC70含量在第5天显著高于治疗组和病毒组(P<0.05);在第7,11天时,各组组内热休克同源蛋白HSC70的含量无明显差异(P>0.05);第9天病毒组的热休克同源蛋白HSC70含量显著高于其他组(P<0.05)。HSC70 mRNA含量随天数的增加而减少,正常组下降趋势显著(P<0.05),预防组在第3,4天趋于平缓。攻RV后小鼠肠内HSC70 mRNA的相对表达量在第5,7,11天,呈现为病毒组>预防组>治疗组>正常组(P<0.05),且小鼠肠内HSC70 mRNA的相对表达量在第7天显著升高(P<0.05)。对第9天空肠组织进行冰冻切片-间接免疫荧光检测发现,HSC70主要存在于空肠绒毛组织,且预防组中HSC70明显少于病毒组及正常组(P<0.05)。结果表明,食淀粉乳杆菌可显著抑制RV受体HSC70在细胞中的合成,从而发挥抗病毒作用,为探究食淀粉乳杆菌抗RV机理提供理论依据。

摘  要:
The aim of this study was to investigate the effect of Lactobacillus amylovorus on the cellular receptor heat shock cognate HSC70 of rotavirus(RV).The Kunming suckling mice fed with Lactobacillus amylovorus were used as experimental animals.The jejunal tissue was collected from the 1~(st) to 11~(th) day and detected HSC70 by ELISA,qPCR,and frozen section indirect immunofluorescence.The test was divided into four groups: normal group,virus group,prevention group(attack RV after probiotics),treatment group(probiotics after RV).The experimental results showed that the growth rate and change of weight of suckling mice were related to the RV infection,and Lactobacillus amylovorus could significantly treat and prevent the change caused by the RV infection(P<0.05).The detection of HSC70 in jejunal tissue showed that the content of HSC70 in the normal group and prevention group was significantly higher than that in the treatment group and virus group(P<0.05).At 7~(th) and 11~(th)day,there was no significant difference in the content of HSC70 in each group(P>0.05).The content of HSC70 in 9~(th) day virus group was significantly higher than the other groups(P<0.05).The level of HSC70 mRNA decreased with the days,the decline trend of the normal group was significant(P<0.05),and the prevention group tended to be gentle at 3~(rd) and 4~(th) day.At 5~(th),7~(th),11~(th) day,the relative expression of HSC70 mRNA in the intestine of mice after RV was the virus group> prevention group> treatment group> normal group(P<0.05),and the relative expression of HSC70 mRNA in the intestine was significantly elevated at 7~(th) day(P<0.05).The indirect immunofluorescence assay of 9~(th) day frozen jejunal tissue showed that HSC70 was mainly found in jejunum villus,and the HSC70 in the prevention group was significantly less than that in the virus group and normal group(P<0.05).The results showed that Lactobacillus amylovorus can significantly inhibit the synthesis of RV receptor HSC70 in cells,thereby exerting antiviral effect,and providing a theoretical basis for exploring the anti-rotavirus mechanism of Lactobacillus amylovorus.
