新疆石河子地区鸡皮刺螨形态学鉴定及其cox1基因比对分析

摘  要:
为了确定新疆石河子地区流行的鸡螨虫的种类,采集本地区蛋鸡和斗鸡流行的螨样进行形态学观察鉴定,同时克隆并分析其线粒体DNA细胞色素C氧化酶亚基1(cox1)基因,并与Gen Bank登录的鸡皮刺螨参考序列对比分析同源性并构建系统进化树。初步形态学鉴定结果显示:该虫体呈长椭圆形,后部略宽,螯枝细而长;背面有1块背板,前部较宽,后缘平直;胸板拱桥形,宽大于长,板上刚毛2对;肛板圆三角形,肛孔位于肛板后部;雄螨的胸殖板和腹肛板相接形成全腹板,具有鸡皮刺螨的形态特征。分子生物学鉴定发现,本研究螨虫株cox1基因与法国和意大利鸡源鸡皮刺螨cox1基因序列同源性达99%,证实石河子地区流行的鸡螨虫为鸡皮刺螨,该螨虫与法国和意大利鸡源鸡皮刺螨具有较近的亲缘关系。

摘  要:
To clarify the species of chicken mites in the regicon of Shihezi, popular chicken mite samples in laying hens and fighting cocks in the region were collected and identified by morphological observation. Then mitochondrial DNA cytochrome C oxidase subunit 1(cox1) gene was cloned and sequenced. The sequences were analyzed by comparison with reference sequence of chicken mite deposited in Gen Bank, and homology and phylogenetic tree were analyzed. The morphological observation of the mites showed that collected chicken mites possess elongated oval, slightly wider rear, chelating branch thin and long; One piece back on the back, a wide front, the rear edge of the flat; chest plate arch shaped, more wide than long, on- board two pairs of bristles; anal plate round triangle, anal orifice plate located in the rear; sternal- genito plate and ventro- anal plate of male reproductive mites contact to form holoventral plate, which were preliminary identified as Dermanyssus gallinae. Identification of molecular biology based on cox1 gene indicated that the species of mites were Dermanyssus gallinae, which shared identities of99% for cox1 gene, with France and Italy chicken mite and had a closer relationship.
