极端低温胁迫野扇花叶片耐寒生理生化特性的研究

摘  要:
为研究野扇花对低温环境的适应特性,实验对野生野扇花不同休眠时期叶片进行极端低温(18℃、0℃、-5℃、-10℃、-15℃)胁迫处理,测试野扇花游离脯氨酸含量、POD酶活性及细胞膜透性等耐寒能力的抗性指标,研究结果表明:三个不同越冬时期游离脯氨酸含量变化规律基本一致,当温度在-5℃~-10℃时含量较高,说明该温度下叶片脯氨酸具有较强的抗寒能力;POD酶活性测定结果表明野扇花在不同休眠期表现出不同的酶活性,且休眠末期活性最强,深休眠期-5℃时具有较好的抵御低温胁迫的能力,随着胁迫强度加深,酶活性减小;在-10℃~-15℃时,其相对电导率值急剧增大,经非线性回归分析、Logistic曲线拟合求出拐点温度(LT50)为-12.73℃。研究结果表明野扇花越冬期具有相应的耐寒能力。

摘  要:
In order to understand the adaptability of Sarcococca ruscifolia to the low temperature environment, the leaves of Sarcococca ruscifolia at different dormant periods were treated with different levels of low temperatures( 18℃,0 ℃,- 5 ℃,- 10 ℃,- 15 ℃),after which the stress resistance indexes were measured including the free proline content,the POD enzyme activity and the cell membrane permeability. The result showed that the changes of free proline content at three different overwintering period were basically the same,with higher value at- 5 ℃ ~- 10 ℃ showing that the plants have a stronger cold resistance at that temperature; POD assay showed different enzymatic activities at different dormant periods with the strongest activity at the late stage,and the plant showed better ability to resist low temperature stress in deep dormancy at- 5 ℃ with decreasing enzymatic activity when the stress intensity deepened; at the temperature of- 10 ℃ ~- 15 ℃,the relative conductivity increased rapidly,and the inflection point temperature( LT50) was- 12. 73 ℃,which was come out by method of nonlinear regression analysis and logistic curve fitting. The results showed that Sarcococca ruscifolia has the corresponding ability to resist cold temperature during overwintering period.
