摘  要:
为考察碳源、培养时间、温度、起始p H等因素对产果胶酶疣孢青霉(Penicillium verruculosum Peyron.)菌株TS63-9发酵产酶的影响,对该菌株发酵条件进行了优化,并考察了最优发酵条件下粗酶液在烟丝中的应用效果。结果表明,TS63-9液体发酵最佳碳源为烟梗粉末,添加量7.5 g/L;最适培养温度为28℃,最适起始p H为6.0;经优化后,在摇床转速200 r/min条件下培养72 h,粗酶液果胶酶活性达到832.49U/m L。将此粗酶液添加应用于烟丝后,可显著降低烟丝果胶含量,感官质量得到一定程度改善;通过扫描电镜观察烟丝表面细胞排列特征,发现细胞间隙变大,细胞间隙比例提高。

摘  要:
Different conditions including carbon source, incubation time, temperature and p H for the production of pectinase by Penicillium verruculosum Peyron. TS63-9 had been studied. Then, the effects of crude enzyme of TS63-9 on cut tobacco were investigated from different areas. The results showed that, the optimium carbon source was 7.5 g / L tobacco stem powder,the optimium temperature and p H was 28 ℃ and 6.0. Under these conditions, the highest pectinase activity reached to 832.49 U / m L after incubating for 72 h at rotating speed 200 r / min. TS63-9 crude enzyme could decrease the pectin content significantly after treating several hours at given conditions. The smoking quality of treated cut tobacco was improved to a certain extent. Cell morphological characteristics of tobacco surface showed that the cell gaps became larger than untreated sample;and the ratios of cell gap increased greatly.
