太空环境诱导的草地早熟禾皱叶突变体蛋白质组学研究

摘  要:
“神舟3号”飞船搭载草地早熟禾(Poa pratensisL.)‘Nassau’干种子,地面种植后筛选出皱叶突变株系PM2。利用蛋白质组学技术对PM2突变株系叶片中表达的蛋白质进行解析(未搭载植株为对照),发现基本保持不变的蛋白质点有181个,上调节表达蛋白质点有15个,下调节表达蛋白质点有15个,失去表达蛋白质点有116个。选择19个蛋白质点进行肽质谱分析,同源性查询后其中10个蛋白质点得到阳性结果,且PM2-058为谷氨酸合成酶类似蛋白,PM2-175为磷酸丙糖异构酶类似蛋白;CK-103为ATP酶依赖型26S蛋白酶体类似蛋白;CK-330为1,5二磷酸核酮糖羧化酶/加氧酶类似蛋白;CK-376为硫氧还蛋白M前体类似蛋白;CK-391为二磷酸核酮糖羧化酶类似蛋白。

摘  要:
The dry seeds of Poa pratensis L.variety 'Nassau' were carried by spaceship SZ-3 as treatments and the seeds which on the earth as CK.A winnkle-leaf mutant which named PM_2 was selected for the analysis.Proteins expressed in leaf of CK and PM_2 were compared and profiled with the techniques of proteomics.There are 181 proteins maintained no changes,15 proteins were up-regulated and 15 proteins were down-regulated,and 116 proteins were lost. 19 proteins were chosen to make pdypeptide fingerprint and homelogy analysis.It indicated that,10 of them were positive results and protein PM_2-058 was close to glutamine synthetase,PM_2-175 was close to triosephosphate isomerase,CK-103 was close to 26S proteasome regulatory complex ATPase,CK-330 was close to ribulose 1,5 biphosphate carboxylase/oxygenase,CK-376 was close to thioredoxin M precursor,CK-391 was close to ribulose bisphosphate carboxylase.
