基于线粒体nad1和nad4基因对四川省多头带绦虫种群遗传多样性的分析

摘  要:
为探讨多头带绦虫的种群遗传多样性,应用PCR方法扩增20株分离自四川省攀枝花市、雅安市、成都市的山羊脑部和肌间的多头蚴的线粒体nad1基因和nad4基因的部分序列,并进行分析。序列分析结果显示,20株多头蚴的pnad1基因和pnad4基因序列分别有2个和8个变异位点,长度分别为753bp和801bp,变异率分别为0~0.3%和0~0.8%,检测出3个和6个单倍型,总的单倍型多样性分别为0.574±0.055和0.742±0.074,核苷酸多样性分别为0.000 82±0.000 53和0.002 88±0.001,平均遗传距离分别为0.002和0.003。构建的种系发育树显示,四川分离株与已知多头带绦虫位于同一分支,且与分布的地理区域、寄生部位没有直接的相关性。结果表明,四川多头带绦虫的遗传多样性水平低,其中nad1基因的遗传多样性水平低于nad4基因的遗传多样性。

摘  要:
The aim of this study was to determine the level of population genetic diversity of Taenia multiceps.Partial sequences of the mitochondrial nad1gene and nad4gene of 20isolates of coenurus collected from the brain or intramuscular of goats in Panzhihua,Ya'an and Chengdu prefectures of Sichuan Province were amplified and analyzed.In result,there were 2variable sites and 8variable sites in the pnad1 and pnad4sequences,the mitochondrial genes sequenced in this study were 753bp for pnad1and 801bp for pnad4in length,and the sequence variations of pnad1gene and pnad4gene were from 0to 0.3%and from 0to 0.8%,respectively.Three haplotypes and 6haplotypes were detected in pnad1gene and pnad4 gene,with the global haplotype diversity being 0.574±0.055(pnad1gene)and 0.742±0.074(pnad4 gene).The nucleotide diversities were 0.000 82±0.000 53for pnad1gene and 0.002 88±0.001for pnad4 gene,and the average genetic distances were 0.002for pnad1gene and 0.003for pnad4gene,respectively. The phylogenetic trees showed that all the 20isolates from Sichuan Province and other T.multiceps isolates available in GenBank were clustered in the same clade,and there was no direct correlation between parasitic sites and geographical regions.The results indicated that the level of genetic diversity of T.multiceps from Sichuan Province was low,and the diversity of nad1gene was lower than that of nad4gene.
