辣椒根尖细胞微核制片方法的探讨

摘  要:
为了建立辣椒根尖微核技术,对辣椒种子萌发条件和辣椒根尖解离时间等环节进行了探索。结果表明:52℃和室温浸泡利于辣椒种子萌发,可以得到状态良好的根尖材料;在60℃1N HCl中解离7 min,细胞分散和染色效果较好;利用辣椒根尖细胞可以获得理想的微核制片。

摘  要:
In order to establish micronucleus technology of pepper root tips,the germination conditions of pepper seeds and the dissociation time of pepper roots were investigated.The results showed that 52℃ treatment and soaked at room temperature were beneficial for germination of pepper seeds,the root tip material in good condition may be acquired;Cell dispersion and dyeing was better when dissociating pepper root tips 7 min at 60℃ with 1N HCl;Using pepper root tips can get ideal micronucleus preparation.
