利用全自动凯氏定氮仪测定大豆粗蛋白含量的条件研究

摘  要:
[目的]研究多种因素对大豆粗蛋白含量测定的影响,以期获得最优化的试验条件.[方法]利用全自动凯氏定氮仪比较不同称样量、催化剂比例、硫酸用量、消化时间、消化温度条件下测定的大豆粗蛋白含量.[结果]当只改变某一种试验条件的水平时,大豆粗蛋白含量测定结果之间差异较大,有的甚至达到极显著水平.针对大豆样本,较优化的试验条件是称样量0.5 g、催化剂用量比例K2 SO4/CuSO4·5H2O为6/0.2(g/g)、硫酸用量为8 mL、消化时间105 min、消化温度420℃.仪器测定硫酸铵的回收率为100.26%,符合定量的要求.[结论]该研究为其他相关研究提供方法借鉴.

摘  要:
[Objective]The research aimed to study the effects of various factors on the determination of soybean crude protein content in order to obtain optimal experimental conditions.[Method]Soybean crude protein content was measured by using automatic kjeldahl analyzer under different conditions, which include sample amount, ratio of catalyst, sulfuric acid amount, digesting time and digesting temperature. [Result]When only the level of a certain test condition was changed, the difference between the determination results of the soybean crude protein content was large, and some even reached a very significant level.For the soybean samples, the optimized experimental conditions were 0.5 g soybean sample, the ratio of catalyst to K_2SO_4/CuSO_4·5 H_2O was 6 g/0.2 g, the amount of sulfuric acid was 8 mL, the digestion time was 105 min, and the digestion temperature was 420 ℃.The recovery of(NH_4)_2SO_4 by using automatic kjeldahl analyzer was 100.26%, which was suitable for quantitative analysis.[Conclusion]This study provides a methodological reference for other related research.
