野生大熊猫粪便源携Ⅰ类整合子的产志贺毒素大肠杆菌的分离鉴定

摘  要:
为了解四川某地区野生大熊猫体内是否携带产志贺毒素大肠杆菌(STEC),本研究采用选择培养基从采集的野生大熊猫粪便中进行细菌分离,结果得到一株β溶血的大肠杆菌。通过菌落形态学观察、革兰染色镜检、生化试验、16S rDNA序列测定分析、毒力基因检测、致病性实验及药敏实验进一步鉴定。结果表明:该菌株属于STEC;接种2×10~8 cfu/0.2 mL剂量该菌的小鼠在24 h内全部死亡,表明该菌株具有较强的致病性;药敏结果显示,该菌株对四环素、头孢噻肟、复方新诺明、卡那霉素、链霉素、哌拉西林、诺氟沙星、强力霉素、头孢呋辛存在多重耐药;该菌株携带Ⅰ类整合子基因。本研究结果对野生大熊猫疫病防控及健康风险评估具有一定的指导意义。

摘  要:
To study whether the wild giant pandas carry Shinga Toxin-producing Escherichia coli(STEC) in Sichuan Province, a beta hemolytic E.coli was isolated from feces collected from wild giant pandas using a selective medium in this study.It was further identified by colony morphology observation, gram staining microscopy, biochemical test, 16 S rDNA sequence analysis, virulence gene detection, pathogenicity test and drug sensitivity test. The results showed that the strain was STEC. Mice inoculated with 2.0 ×10~8 cfu/0.2 mL doses of this strain died within 24 h, indicating that the strain had high pathogenicity. The drug sensitive test suggested that this strain had multi-drug resistance to tetracycline, cefotaxime, cotrimoxazole, kanamycin,streptomycin, piperacillin, norfloxacin, doxycycline and cefuroxime, and carried class Ⅰ integron genes. This study had some guiding significance for prevention and health risk assessment of wild giant panda.
