甘草查尔酮A抑制小鼠黑色素瘤B16F10细胞糖酵解表型和诱导凋亡

摘  要:
为探讨甘草查尔酮A对小鼠黑色素瘤B16F10细胞糖酵解表型和凋亡的影响。采用磺酰罗丹明B法检测甘草查尔酮A对B16F10细胞增殖影响的方法,比色法检测B16F10细胞外葡萄糖和乳酸含量,生物发光法检测细胞内ATP含量,Giemsa和Hoechst33258染色法观察细胞形态变化,Annexin V-FITC/PI双染法检测细胞凋亡率。结果显示,甘草查尔酮A能有效抑制B16F10细胞增殖,呈现浓度依赖性和时间依赖性。随药物浓度的增加,B16F10细胞摄取葡萄糖能力减弱,细胞外乳酸含量和细胞内ATP含量明显降低。同时,细胞出现典型凋亡形态,细胞体积逐渐缩小,胞核皱缩,碎裂,呈现致密颗粒状荧光,且细胞凋亡率明显增加。由此可知,甘草查尔酮A能抑制B16F10细胞糖酵解表型和诱导细胞凋亡。

摘  要:
To investigate the effects of licochalcone A on the glycolytic phenotype and apoptosis in melanoma B16F10 cells. The proliferation of B16F10 cells induced by Licochalcone A was determined by Sulforhodamine B(SRB) method. The levels of extracellular glucose and lactic acid were detected by chromatometry, and intracellular ATP levels were measured by bioluminescence assay. The cellular morphological changes were observed by Giemsa and Hoechst 33258 staining. The apoptosis rate was determined by Annexin V-FITC/PI assay. The proliferation of B16F10 cells treated with Licochalcone A was effectively inhibited in a concentration and time-dependent manner. A substantial decrease in glucose uptake, extracellular lactic acid and intracellular ATP levels were observed. The typical apoptosis morphology(nuclear condensation and fragmentation) was observed with the increasing concentration of Licochalcone A in B16F10 cells, which was associated with the increasing apoptotic rates.It was concluded that the licochalcone A can inhibit glycolytic phenotype and induce apoptosis in melanoma B16F10 cells.
