摘  要:
【目的】探讨牦牛血中IgG的最佳提取工艺.【方法】以牦牛血液为原料,在单因素试验的基础上,选取聚乙二醇浓度、K2HPO4浓度、NaCl浓度和pH为自变量,IgG的含量为响应值,根据响应面Box-Behnken试验设计原理,采用四因子三水平的分析法模拟得到二次多项式回归方程的预测模型.优化聚乙二醇/K2HPO4双水相法提取牦牛血中IgG的工艺条件.【结果】聚乙二醇/K2HPO4双水相法提取牦牛血中IgG的最佳工艺条件为聚乙二醇13%(w/w)、K2HPO417%(w/w)、NaCl 11%(w/w)、pH 6.0,在此条件下IgG质量浓度达13.82mg/mL,与预测值14.04mg/mL的相对误差为1.59%.【结论】回归模型具有高度显著性,方程对试验拟合较好,可以对IgG的含量进行很好的分析和预测;各因子对IgG含量影响大小依次是PEG浓度>NaCl浓度>K2HPO4浓度>pH;

摘  要:
【Objective】To optimize the extraction process for IgG from yak blood.【Method】Yak blood was used as raw material,the extraction condition was optimized on the basis of single-factor test and the response surface methodology,IgG was extracted from yak blood with PEG/K2HPO4 aqueous two-phase system.【Result】The optimum extraction parameters were as PEG 13%(w/w),K2HPO417%(w/w),NaCl 11%(w/w),pH 6.0.The IgG content reached to 13.82 mg/mL under the conditions above mentioned,and the relative error was 1.59% with the theoretical value at 14.04 mg/mL.【Conclusion】The model has a significant difference,and the established regression equation for the concentration of IgG has an excellent goodness of fit.The concentration of IgG could be analyzed and predicted by the model.Factors influencing the extraction are in the order as:PEG concentration>NaCl concentration>K2HPO4concentration>pH.
