7种中药材斑蝥COI基因序列的分子系统学研究

摘  要:
以我国重要的中药资源芫菁科7种斑蝥为研究对象,分析线粒体COI基因序列,探讨COI基因作为DNA条形码在鉴定药用斑蝥基源方面的可行性。采用MEGA 5.0软件分析了不同种之间的碱基组成、遗传距离,利用邻位相连法(NJ)和最大简约法(MP)分别构建了系统发育树。结果显示,PCR扩增获得长约700 bp,GC含量为37.8%~41.4%的COI基因片段。7种斑蝥种间遗传距离较大,介于0.10~0.23之间。系统进化树显示,不同属各自聚为一支,同属不同物种的个体也形成各自独立的分支,基于COI基因的DNA条形码在识别芫菁物种方面和形态学一致。本研究表明以COI基因作为芫菁DNA条形码进行物种鉴定具有一定的可行性。

摘  要:
In this paper,taken seven Chinese blister beetle species as tested objects,their sequences of mitochondrial COI were analyzed,and the viability of the COI gene for DNA barcoding and species identification were also studied. The DNA base composition and genetic distance among the seven species were calculated by using MEGA 5. 0,and the phylogenetic tree were constructed with Neighbor-Joining(NJ)and Maximum Parsimony(MP) methods. The results showed that a 700 bp gene fragment was obtained by using PCR amplification,which GC contents was in the range from 37. 8 % to 41. 4 %. Genetic distance of the seven Chinese blister beetle ranged from 0. 10 to 0. 23. The seven Chinese blister beetle could be easily divided to different branchs by NJ tree and MP tree. Species identification with COI sequences in Chinese blister beetle supported the taxonomy based on morphological characters. The conclusion of our research suggested that mitochondrial COI gene was a valid DNA barcoding gene for specie identification in Meloidae.
