大麦MBF1基因的电子克隆与生物信息学分析

摘  要:
通过同源比对,对大麦MBF1s基因进行了电子克隆,并通过生物信息学的方法对其及编码蛋白质进行了预测和分析。结果表明,克隆到大麦MBF1家族3个成员Hv MBF1a、Hv MBF1b和Hv MBF1c,三者所编码蛋白质均包含典型的MBF1和HLH结构域,无可识别的信号肽和跨膜区,均为亲水性蛋白,含有数个可能的磷酸化位点。电子表达谱结果表明,3个成员在大麦的大部分组织器官中都有表达,而Hv MBF1a和Hv MBF1b对所分析的胁迫处理响应不明显,Hv MBF1c的表达受各种处理的剧烈诱导,Hv MBF1c可以作为大麦和其他作物抗逆性分子育种的重要候选基因。

摘  要:
Electronic cloning and bioinformatics analysis of MBF1 genes were studied and putative proteins were coded by them from barley. The results showed that three members of MBF1 gene family were cloned from barley,namely Hv MBF1 a,Hv MBF1 b and Hv MBF1 c. All of the three MBF1 proteins contained typical MBF1 and HLH conserved domains at N-and C-terminal,respectively.There was no recognized signal peptide or transmembrane region in these proteins.All of them were hydrophilic proteins,and harbored several phosphorylation sites. Silico analysis showed that the expression of the three Hv MBF1 s could be detected in most of the organs of barley,and the expression of Hv MBF1 a and Hv MBF1 b was not influenced by various stimuluses,while Hv MBF1 c showed quite different expression level under biotic and abiotic stresses,which could be used as an important candidate gene in molecular breeding of barley and other crops with improved stress tolerance.
