小粒黑豆GmRHF1基因生物信息学及线虫诱导表达分析

摘  要:
为研究E3泛素连接酶基因在大豆抗线虫病害中的功能,利用RT-PCR方法从大豆胞囊线虫(Heterodera glycines)侵染的小粒黑豆(Glycine max)根部组织中克隆到1个E3泛素连接酶RING家族基因GmRHF1.序列分析结果显示,该基因位于大豆基因组3号染色体上,具有一个完整的外显子,编码区长度783 bp,编码260个氨基酸,含有1个RING-H2模体和1个跨膜结构域.蛋白质分子质量为27 880.43 u,理论等电点为5.59.同源性比对及系统进化分析表明,该蛋白质与ATL3蛋白亲缘关系最近,属于RING-H2 zinc finger ATL3类.实时荧光定量PCR(qRT-PCR)分析GmRHF1受大豆胞囊线虫侵染的诱导表达情况,结果表明,接种后48 h内该基因的表达量较未接种的样品显著增加,推测该基因介导小粒黑豆抗大豆胞囊线虫的防御反应.

摘  要:
In order to study the function of E3 ubiquitin ligase gene in soybean resistance to plant nematode,a gene named GmRHF1 encoding RING type in E3 ubiquitin ligase was cloned from small black bean(Glycine max)roots infected by Heterodera glycines by RT-PCR method.Sequence analysis showed that the gene was located at G.max chromosome 3,and included an exon.The CDS length was 783 bp,encoding 260 amino acid residues.The GmRHF1 protein contained one RING-H2 motif and one transmembrane domain,its molecular weight was 27 880.43 u,and the theoretical isoelectric point was 5.59.The homology alignment and phylogenetic analysis showed that the GmRHF1 protein was closely related to ATL3,classified into RING-H2 zinc finger ATL3 class.Real time PCR analysis revealed that GmRHF1 mRNA levels were significantly increased after exposure to H.glycines from 12 h to 48 h compared with the control without inoculation,which suggested that GmRHF1 was involved in defense response of small black bean to H.glycines.
