乙醇辅助盐析分离猪胃蛋白酶

摘  要:
为确定盐析分离猪胃蛋白酶的最佳分离工艺条件,以猪胃粘膜为原料,经盐酸浸提,通过乙醇辅助盐析对照试验、透析冷冻干燥得猪胃蛋白酶比活力为102927.5U·g-1,酶经fSephadex-G75洗脱得到单一蛋白峰,再经SDS-聚丙烯酰胺凝胶电泳,表现为单一蛋白,酶的亚基相对分子质量约为35ku。

摘  要:
To optimize the technology of the swine pepsin separation,the swine gastric mucosa was extracted by hydrochloric acid.And the crude enzyme was purified through salting-out,dialyzing,freezing drying and sephadex G-75 gel filtration.The purified enzyme was demonstrated by SDS-PAGE to be a homogeneous protein,its molecular mass was 35 ku and the activity was 102 927.5 U·g-1.
