化学诱变剂对同源四倍体毛泡桐体外植株再生的影响

摘  要:
在建立同源四倍体毛泡桐体外植株再生系统的同时,研究了甲基磺酸甲酯(MMS,DNA甲基剂)和5-氮胞苷(5-Aza,DNA去甲基剂)对以叶片为外植体的体外植株再生系统的影响.结果表明,同源四倍体毛泡桐幼苗叶片在无MMS和5-Aza的MS培养基上,愈伤组织最高诱导率皆可达到100%,而幼苗叶柄和茎段则只能在NAA 0.7,0.9,1.1 mg.L-1,BA 2和4 mg.L-1组合的MS培养基上达到100%.叶片愈伤组织芽诱导率达到100%的培养基为MS+NAA 0.9 mg.L-1+BA 16 mg.L-1.MMS和5-Aza对同源四倍体毛泡桐叶片体外植株再生有抑制作用,并且随着MMS和5-Aza质量浓度的升高,抑制作用越明显.此外,5-Aza对叶片芽和根诱导的抑制作用皆大于MMS.

摘  要:
A study was conducted on the effect of chemical mutagens on in vitro plantlet regenerations from the leaves of Paulownia tomentosa autotetraploid,based on the establishment of system of in vitro plantlet regeneration from the different explants of the autotetraploid.The results indicated that the highest callus induction rate of the leaves could reach 100% on MS media supplemented with NAA and BA and free MMS and 5-Aza,but the petiole and stem segments only on the media with NAA 0.7,0.9,1.1 mg·L-1 and BA 2-4 mg·L-1.The medium on which the shoot induction rate of leaf callus could reach 100% was MS+ NAA 0.9 mg·L-1+BA 16 mg·L-1.MMS and 5-Aza might inhibit the in vitro plantlet regenerations of the autotetraploid leaves,and the inhibitory effect was greater with the increase of MMS and 5-Aza concentration.Moreover,the inhibitory effect of 5-Aza on the shoot and root induction was greater than that of MMS.
