长白楤木腋芽愈伤组织诱导及植株再生

摘  要:
为了在短时间内获得充足的长白楤木苗木,以腋芽为材料进行了其愈伤组织诱导和植株再生研究。结果表明,长白楤木腋芽愈伤组织诱导最佳培养基为MS+BA 1.5 mg/L+2,4-D 1.0 mg/L,诱导率达100%;最佳不定芽分化培养基为MS+BA 2.0 mg/L+NAA 0.2mg/L+IAA 0.2 mg/L,分化率可达90%;最佳不定芽生根培养基为1/2MS+NAA 0.02 mg/L和1/2MS+IBA 0.5 mg/L,生根率达100%。经3~5d炼苗的植株移栽效果较为理想。生根幼株在落叶松腐殖土加河沙和落叶松腐殖土加蛭石配比的2种基质上移栽效果较好。

摘  要:
In order to obtain plenty of Aralia continentalis Kitag.seedlings in a short time,callus induction and plant regeneration was conducted by taking axillary buds as explants.The results showed that the optimum medium for callus induction from A.continentalis axillary bud was MS supplemented with 1.5 mg/L BA and 1.0 mg/L 2,4-D,and the induction rate was up to 100%.The optimum medium for adventitious shoot differentiation was MS supplemented with 2.0 mg/L BA,0.2 mg/L NAA and 0.2 mg/L IAA,and the differentiation rate was 90%.The optimum medium for shoot rooting was 1/2 MS supplemented with 0.02 mg/L NAA or 0.5 mg/L IBA,and the rooting rate was 100%.The plantlets should be transplanted after acclimatization for 3-5 days.The better transplanting effect was obtained with the growing media composed by larch forest humus mixed with sand or vermiculite.
