金桔体细胞胚胎诱导的研究

摘  要：
采用金桔无菌苗叶片为外植体,在单因素试验的基础上,通过正交试验研究不同生长调节剂对金桔胚性愈伤组织诱导的影响。结果表明:MS+2,4-D 2.0mg·L-1+BA 0.5mg·L-1处理最适宜金桔胚性愈伤组织诱导,其中淡黄色愈伤组织团块胚胎发生频率较高;在附加BA 0.05mg·L-1的MS培养基上淡黄色愈伤组织块可形成胚状体;胚状体试管苗在1/2MS+NAA0.1mg·L-1培养基上,生根率可达80%,移栽成活率达70%

摘  要：
Based on single factorial experiment,the sterile seedlings of Fortunella japonica were used as explants for studying the effects of various growth regulators on inducing embryogeny with orthogonal test.The results showed:the MS+2,4-D 2.0mg·L-1+BA 0.5mg·L-1 was the best culture medium for inducing embryonic callus.Sucrose at MS culture medium with BA 0.05mg·L-1 gave a maximum embryogenesis from the light yellow callus tissue of leaf.Planting somatic embryos on 1/2MS culture medium with NAA0.1mg·L-1,the rooting rate was 80%,the survive rate was 70%.
