基于matK基因的几种苔藓植物的亲缘关系分析

摘  要:
以钝鳞紫背苔(Plagiochasma appendiculation)为外类群(outgroup),利用ClustalX 2.0和MEGA 4.1软件对8种苔藓植物的matK基因序列进行比对和分析,构建分子系统树。结果表明:8种苔藓植物的matK序列长度为638bp~650bp,排序后总长度为664bp,其中变异位点357个,信息位点180个。遗传距离为0.058~0.557,平均遗传距离为0.287。利用NJ法建立的系统树显示,8种苔藓植物分为3组,其中2种丛藓科的聚为一组,3种青藓科的聚为一组,3种灰藓科的聚为另一组。序列分析结果与形态学结果一致,表明matK基因序列可用于苔藓植物的亲缘关系分析。

摘  要:
Using Plagiochasma appendiculation as the outgroup,matK genes belonging to 8 bryophyte species were analyzed and compared to construct the phylogenetic trees with ClustalX 2.0 and MEGA 4.1 softwares.The results showed that the length of matK gene in 8 bryophytes were between 638 bp and 650 bp and the total length of sequence was 664 bp after alignment,including 357 variable sites and 180 informative sites.The genetic distance were 0.058-0.557 averaging 0.287.The 8 species in the NJ tree(neighbor-joining tree) were clustered into three groups: 2 species of Pottiaceae,3 species of Brachytheciaceae and 3 species of Hypnaceae.The clustered results were consistent with the morphologic identification,which also showed that matK gene can be used for the analysis of bryophytes relationship.
