桉树枝瘿姬小蜂雌成虫体内可培养细菌分离鉴定及多样性分析

摘  要:
【目的】明确桉树枝瘿姬小蜂(Leptocybe invasa Fisher&La Salle)体内可培养细菌多样性,为利用微生物防治该害虫打下基础。【方法】利用传统的细菌分离培养与鉴定方法,结合16S rDNA测序和系统发育分析,对刚羽化的桉树枝瘿姬小蜂雌成虫体内可培养细菌进行多样性分析。【结果】从桉树枝瘿姬小蜂体内分离出11株细菌,经鉴定归类于3个门(厚壁菌门、放线菌门和变形菌门)3个纲(芽孢杆菌纲、放线菌纲和γ-变形菌纲)4个目(芽孢杆菌目、微球菌目、乳酸杆菌目和肠杆菌目)8个科(芽孢杆菌科、微杆菌科、葡萄球菌科、乳酸杆菌科、纤维单胞菌科、微球菌科、肠杆菌科和类芽孢杆菌科)8个属(芽孢杆菌属、微杆菌属、葡萄球菌属、片球菌属、纤维单胞菌属、微球菌属、埃希氏杆菌属和类芽孢杆菌属),其中,厚壁菌门(Firmicutes)6株,为优势类群;芽孢杆菌属(Bacillus)3株,为优势菌属。【结论】桉树枝瘿姬小蜂体内可培养细菌具有丰富的多样性。

摘  要:
【Objective】The diversity of culturable bacteria in Leptocybe invasa Fisher & La Salle was clarified to lay a foundation for microbiological control on the pest.【Method】Traditional methods of bacterial isolation,culture and identification combined with 16 S rDNA sequence and phylogenetic analysis were used for the study on the diversity of culturable bacteria in the female adults of L.invasa that were newly emerged.【Result】Eleven strains of bacteria were isolated from the sexually matured female L.invasa,belonging to three phyla(Firmicutes,Actinobacteria and Proteobacteria),three classes(Bacilli,Actinobacteria and Gammaproteobacteria),four orders(Bacillales,Micrococcales,Lactobacillales and Enterobacterales),eight families(Bacillacea,Microbacteriaceae,Staphylococcaceae,Lactobacillaceae,Cellulomonadaceae,Micrococcaceae,Enterobacteriaceae and Paenibacillaceae)and eight genera(Bacillus,Microbacterium,Staphylococcus,Pediococcus,Cellulomonas,Microbacterium,Escherichia and Paenibacillus),among which Firmicutes(six strains)was the dominant bacterium and Bacillus(three strains)was the dominant genus.【Conclusion】The diversity of culturable bacteria in L.invasa is rich.
