急性冷暴露对仔猪比目鱼肌GSK3β及凋亡蛋白Caspase-3表达的影响

摘  要:
气候条件与畜牧业的发展密切相关,尤其在我国北方寒区,养殖动物极易受到寒冷刺激的危害,但寒冷刺激引起动物机体损伤的机制尚不明确,因此本研究旨在探究急性冷暴露对仔猪比目鱼肌糖原合酶激酶3β(GSK3β)及凋亡蛋白半胱天冬酶-3(Caspase-3)表达的影响。将12只仔猪随机分成3组,分别为常温对照组、冷暴露4 h组和冷暴露8 h组,采集各组仔猪比目鱼肌样本进行Western blot分析。结果显示,冷暴露组Cleaved Caspase-3和GSK3β磷酸化显著高于对照组,B淋巴细胞瘤-2基因(Bcl-2)/Bcl-2-Associated X蛋白质(Bax)比值与对照组相比显著降低。结果表明,急性冷暴露引起Cleaved Caspase-3和GSK3β磷酸化水平的升高和Bcl-2/Bax比值的降低,推测可能是机体通过抑制GSK3β的磷酸化来减弱急性冷暴露诱导的细胞凋亡。

摘  要:
The climatic conditions are closely related to the development of animal husbandry.Especially in the cold regions of Northern China,farmed animals are highly vulnerable to cold stimuli,but the mechanism of cold stimuli-induced damage to animal organisms is still unclear.Therefore,the present study aimed to investigate the effects of acute cold exposure on the expression of glycogen synthase kinase 3β(GSK3β)and Caspase-3 in the soleus muscle of piglets.12 piglets were randomly divided into three groups:normal temperature control group,cold exposure 4 h group and cold exposure 8 h group.Western blot analysis was performed for detecting the soleus muscle samples of the piglets.The results showed that GSK3β phosphorylation and Cleaved Caspase-3 expression were significantly higher in the cold exposed group than in the control group,and the Bcl-2/Bax ratio was significantly lower than that in the control group.The results indicated that acute cold exposure increased the level of GSK3β phosphorylation and Cleaved Caspase-3 expression,and decreased the ratio of Bcl-2/Bax.It is speculated that the body may attenuate the apoptosis induced by acute cold exposure by inhibiting the phosphorylation of GSK3β.
