桑树SSR引物的开发及其多态性分析

摘  要:
利用FIASCO磁珠富集法开发出桑树(Morus alba L.)基因组DNA的SSR位点引物46对,选出38对引物用于8份材料的多态性验证。结果表明,38对SSR引物共扩增出清晰谱带210条,不同引物的扩增条带数1~12条,平均每对引物的扩增带数为5.526 3条;每对引物组合可检测到SSR多样性位点数3~9个,共检测到106个多样性位点;所有位点的平均PIC值为0.645 8;8份供试材料在SSR水平上表现出明显的多态性差异,其中野生品种葫芦桑和岩桑聚为Ⅰ组,遗传距离为0.746 0,栽培品种垂桑、荷叶白、剑持、新一之濑、伦教40、鸡桑聚为Ⅱ组,剑持与荷叶白遗传距离最近(0.169 5),亲缘关系最近,葫芦桑和岩桑与荷叶白的亲缘关系最远。

摘  要:
The 46 pairs of SSR sites primers of mulberry genome DNA were developed by FIASCO magnesphere,38 pairs of primers were selected for polymorphic verification of eight materials,the result showed that,210 bands were amplified by 38 pairs of SSR primers clearly. The number of amplified bands were from 1 to 12 with different primers,average amplified bands of each primer were 5.526 3. 3~9 diversity sites of SSR could be detected by each pair of primers,106 diversity sites were detected,The average PIC value was 0.645 8 for all sites. 8 accessions showed obvious polymorphism differences in SSR level. The wild mulberry varieties Hulusang and Yansang were divided into groupsⅠ, the genetic distance was 0.746 0. Cultivated mulberry varieties Cuisang, Heyebai,Jianchi, Xinyizhilai,Lunjiao 40 and Jisang were clustered into group Ⅱ. The genetic distance of Jianchi and Heyebai was the nearest(0.169 5), so the relationship was the closest. The genetic relationship between Hulusang, Yansang and Heyebai were the fatherest.
