摘 要:
为了确定引起广东省江门市某规模化猪场仔猪死亡的原因,试验采集病猪心脏、脾脏、肺脏、关节液、脑、淋巴结等组织进行病毒检测和细菌分离鉴定,并对分离菌进行纯化培养、革兰氏染色、生化鉴定、菌种鉴定及测序分析、血清型鉴定、毒力基因检测、动物试验及药敏试验。结果表明:分离株在鲜血营养琼脂上生长良好,呈α溶血,镜检可见革兰氏阳性球菌,呈典型链状排列;生化特性与猪链球菌相符;PCR扩增得到大小为1 500 bp和688 bp的目的条带,分离株与猪链球菌同源性大于99%,在系统进化树上与猪链球菌处于同一分支,为16型猪链球菌;该分离菌毒力因子基因型为fbps~+/orf~+/sly~+/gapdh~+/epf~-/mrp~-,可致NIH小鼠死亡,毒力较强;分离菌对阿莫西林/克拉维酸、青霉素、头孢噻肟、头孢拉定、头孢曲松、庆大霉素、大观霉素、恩诺沙星和氟苯尼考9种药物敏感,对新霉素、复方磺胺甲口恶唑、强力霉素和替米考星4种药物耐药。选用阿莫西林进行群体治疗,取得了很好的效果。说明该猪场仔猪感染了16型猪链球菌,青霉素类抗菌药物可用于临床治疗。

摘 要:
In order to determine the cause of piglet death in a large-scale pig farm in Jiangmen City, Guangdong Province, the experiment co-llected the hearts, spleens, lungs, synovial fluids, brains, lymph nodes and other tissues of sick pigs for virus detection, bacterial isolation and identification. Purification culture, Gram staining observation, biochemical identification, bacterial species identification and sequencing analysis, serotype identification, virulence gene detection, animal test and drug sensitivity test were carried out on the isolated bacteria. The results showed that the isolated strain grew well on blood agar and showed α-hemolysis; gram-positive cocci were observed by microscopy, which showed typical chain arrangement; the biochemical characteristics were consistent with Streptococcus suis; target bands with sizes of 1 500 bp and 688 bp were obtained by PCR amplification, and the homology between the strain and Streptococcus suis was more than 99%, which was in the same branch with Streptococcus suis in phylogenetic tree, being Streptococcus suis type 16. The virulence factor genotype of the strain was fbps~+/orf~+/sly~+/gapdh~+/epf~-/mrp~-, which could cause the death of NIH mice and have strong virulence. The strain was sensitive to 9 drugs, amoxicillin/clavulanic acid, penicillin, cefotaxime, cefradine, ceftriaxone, gentamicin, spectinomycin, enrofloxacin and florfenicol, and was resistant to four drugs, neomycin, compound sulfamethazole, doxycycline and tilmicosin. Amoxicillin was used for group therapy and achieved good results. The results suggested that the piglets in this pig farm were infected with Streptococcus suis type 16, and penicillin antibacterial drugs could be used for clinical treatment.
