摘 要:
为了挖掘新的抗小麦叶锈病基因,以被无毒小麦叶锈菌诱导的TcLr19为研究对象,通过cDNA末端快速扩增(rapid amplification of cDNA ends,RACE)技术在TcLr19中克隆到抗病相关基因TaABCF,并对其进行表达分析。序列分析表明,TaABCF基因的DNA和cDNA序列长度分别为3 100bp和1 885bp,包含4个内含子和5个外显子,含有两个核苷酸结合域,具有WalkerA、WalkerB和WalkerC保守序列。系统发育分析表明,TaABCF基因与来自乌拉尔图小麦的EMS53714.1基因亲缘关系最近。实时荧光定量分析表明,TaABCF基因在小麦TcLr19与小麦叶锈菌FHPL非亲和互作前期表达提高,而在感病小麦Thatcher与小麦叶锈菌FHPL的亲和互作后期表达受抑制,且在脱落酸(ABA)和茉莉酸甲酯(MeJA)处理下的表达早于水杨酸(SA)处理,说明TaABCF基因参与小麦的抗叶锈反应,且对ABA和MeJA的响应要早于SA。

摘 要:
In order to discover new genes against wheat leaf rust,this study was carried out to clone resistance related genes TaABCFby RACE(rapid amplification of cDNA ends)technique from TcLr19 infected by a virulent Puccinia triticina,and its expression analysis was analysed.The full length cDNA and genomic DNA of the TaABCFgene are 1 885 bp and 3 100 bp,respectively.And it contains 4 introns and 5 exons,and has two nucleotide binding domains and each contains Walker A,Walker B and Walker C.The phylogenetic analyses showed that TaABCF has the closest relationship with EMS53714.1 gene from Triticum urartu Thum.ex Gandil.Expression analysis with real-time fluorescent quantitative PCR showed that the expression of TaABCFwas improved in the incompatible interaction between TcLr19 and P.triticina FHPL in the early stage,while inhibited in the compatible interaction between Thatcher and FHPL latterly.The expression of TaABCF were earlier when treated with ABA and MeJA than that of SA,which showed that TaABCFgene is involved in the resistance of wheat leaf rust and it responds to ABA and MeJA earlier than SA.
