摘  要:
为建立早期快速检测非洲马瘟抗体的IgM捕获ELISA(MAC-ELISA)方法,本试验利用大肠杆菌表达出具有良好抗原性的VP7蛋白,并制备该蛋白的多克隆抗体,通过优化抗原浓度、血清稀释度及酶标抗体稀释度等,建立了检测非洲马瘟的MAC-ELISA方法,并进行了初步应用。结果表明,利用重组表达的VP7蛋白成功建立了检测非洲马瘟的MAC-ELISA方法。

摘  要:
The study was aimed to develop an IgM capture enzyme-linked immunosorbent assay for detecting African horse sickness(AHS). The AHSV VP7 protein with good immunogenicity was expressed in BL21 and polyclonal antibody against protein VP7 of African horse sickness virus was prepared. An IgM capture enzyme-linked immunosorbent assay for detecting AHS was developed by optimizing the antigen concentration,the second antibody concentration and the serum dilution. Preliminary application of the MAC-ELISA method indicated that the MAC-ELISA with the recombined VP7 protein was successfully developed for detecting antibodies to AHSV with good sensitivity and specifi city.
