摘  要:
建立采用紫外光谱技术快速鉴别不同产地美味牛肝菌(Boletus edulis)的方法,确定美味牛肝菌最佳提取溶剂和时间,制备测试液,应用紫外光谱技术建立7个不同产地美味牛肝菌菌盖和菌柄的紫外指纹图谱,光谱数据转化后进行聚类分析。结果显示,其最佳提取溶剂为氯仿,提取时间为30 min;在10 h内的稳定性、方法重现性和精密度的RSD分别在0.04%~1.73%、0.03%~0.63%、0~2.87%之间,表明该方法稳定可靠;菌盖、菌柄的紫外指纹图谱出峰位置相似,而峰高具有差异,表明不同产地美味牛肝菌化学组分相似,含量存在差异;聚类分析结果显示采自同一地区的美味牛肝菌能够很好聚类。研究表明,紫外光谱结合聚类分析能快速鉴别不同产地美味牛肝菌,可作为野生食用菌的鉴别和质量控制新方法。

摘  要:
An ultraviolet spectrum technology was used to rapidly identify Boletus edulis from different regions. For preparing the extracts of B. edulis samples, the best extraction reagent and the best extraction time were determined. The ultraviolet fingerprints of the caps and stipes of B. edulis collected from seven different areas were established with ultraviolet spectrum technology. The spectroscopy data were transposed for the clustering analysis. The results showed as follows : 1 The best extraction reagent of B. edulis was chloroform and the best extraction time was 30 mins. 2 The RSDs(%) of stability,accuracy and repeatability of B. edulis extract within 10 h were 0.04 ~1.73,0 ~2.87 and 0.03 ~0.63 respectively, showed that the method was stable and reliable. 3 The peak positions in the ultraviolet fingerprint of caps and stipes of B. edulis were similar, but the peak heights were different, which showed that the chemical compositions of B. edulis from different regions were similar, but the content was different. 4 The results of cluster analysis showed that B. edulis collected from the same area could have a good clustering. The research concluded that the ultraviolet spectroscopy combined with clustering analysis could provide an effective method for the rapid identification of B. edulis from different regions, as well as provide a new method for discrimination and quality control of wild edible fungi.
