响应面法优选荧光测定维生素C的工艺条件研究

摘  要:
[目的]利用苯甲酸与维生素C反应生成荧光物质的特性,建立一种测量维生素C含量的方法。[方法]试验采用氢氧化钠-邻苯二甲酸氢钾作为缓冲溶液调节缓冲液p H,并通过对测定前处理样品的时间、温度、缓冲液p H以及苯甲酸用量等单因素试验来确定最佳单因素水平,并用响应面法优化试验条件,确定以苯甲酸为促荧光物质测定饮品中维生素C的最佳测定条件。[结果]苯甲酸对维生素C促荧光明显,可在激发波长310 nm,发射波长410 nm处测其产物的荧光强度。维生素C浓度在1.00×10-4~5.05×10-4g/ml范围内其荧光强度呈良好的线性关系,检测限较低,相关系数为0.989。[结论]研究表明,荧光法测定饮品中维生素C结果比较准确,方法可行。

摘  要:
[Objective] Accoding to characteristics of fluorescent substance produced by reaction of Benzoic acid and vitamin C,a method for measuring vitamin C content was established. [Method]Using sodium hydroxide-potassium hydrogen phthalate as buffer solution to adjust p H,through the measurement of sample pre-treatment time,temperature,amount of acid buffer solution and p H test to determine the best univariate single factor levels and response surface optimization of the experimental method to determine the optimum conditions to promote benzoic acid as a fluorescent substance in vitamin C drinks. [Result]Benzoic acid to vitamin C to promote fluorescence evident in the fluorescence intensity of the excitation wavelength of 310 nm,410 nm emission wavelength measured its products. Vitamin C concentration in the 1. 00 × 10- 4~ 5. 05 × 10- 4g / ml scope of its fluorescence intensity was good linear relationship,the lower limit of detection,the correlation coefficient was0. 989. [Conclusion] The result of determining vitamin C content in drink is accurate by fluorescence,the method is feasible.
